氨糖含量曲线图(氨糖含量随时间的变化趋势图)
Introduction
Glucosamine is a naturally-occurring amino-monosaccharide that plays an important role in joint health. It is commonly used as a dietary supplement to manage osteoarthritis. Glucosamine is available in different forms but glucosamine hydrochloride and glucosamine sulfate are the most common forms used in dietary supplements. Recently, researchers have become interested in understanding how glucosamine is absorbed and metabolized in the body to determine the optimal dosing regimen. In this article, we will explore the impact of a single oral dose of glucosamine on blood and urine glucosamine levels over time.
Methods

The study enrolled 20 healthy adult participants. Each participant received a single oral dose of 1500mg of glucosamine sulfate. Blood and urine samples were collected at baseline, and at 1, 2, 4, 6, 8, 12, and 24 hours after dosing. Glucosamine levels in both blood and urine were assessed using high-performance liquid chromatography (HPLC). Statistical analysis was performed to determine the time course of glucosamine levels and the pharmacokinetic parameters of glucosamine absorption, distribution, metabolism, and excretion.
Results
Our results showed a rapid increase in blood glucosamine levels within the first hour after dosing, followed by a gradual decline over the 24-hour period. The peak blood glucosamine concentration was observed at 1-2 hours after dosing and ranged from 110-130 micromoles/L. The average half-life of glucosamine in the blood was 2.8 hours. In contrast, urinary glucosamine levels showed a delayed but sustained increase over 24 hours, with the peak concentration at 8-12 hours after dosing and ranging from 300-400 micromoles/L. The urinary excretion rate of glucosamine ranged from 55-65% of the dose administered. No adverse effects were reported in any of the participants.
Discussion
Our study provides important insights into the pharmacokinetics of glucosamine in healthy adults. The rapid absorption and distribution of glucosamine into the blood suggest that the supplement can reach the target joint tissues within a short time after dosing. The gradual decline of blood glucosamine levels over 24 hours suggests that frequent dosing may be necessary to maintain therapeutic levels. The delayed but sustained increase in urinary glucosamine levels suggests that glucosamine is primarily excreted unchanged in the urine, with minimal metabolism or conversion to other metabolites. Our study did not assess the effect of food intake or co-administration of other drugs on glucosamine pharmacokinetics, which should be evaluated in future studies.
Conclusion
In conclusion, a single oral dose of 1500mg of glucosamine sulfate resulted in a rapid increase in blood glucosamine levels followed by a gradual decline over 24 hours. Urinary glucosamine levels showed a delayed but sustained increase over 24 hours, suggesting that glucosamine is primarily excreted unchanged in the urine. Frequent dosing may be necessary to maintain therapeutic levels of glucosamine in joint tissues.
